Syfte

Although life of thousands of people is saved by reconstructive surgeons many people are still waiting organ donation. Reconstructive surgery became in last decade new discipline -tissue engineering with the focus on the fabrication of living replacement part for the body in the laboratory. Tissue engineering includes biology, chemistry, material science, engineering, immunology and transplantation. Tissue Engineering course provides a general understanding of tissue growth and development, the tools and theoretical information needed to design tissues and organs.

Lärandemål (efter fullgjord kurs ska studenten kunna)

Theoretical objectives



  • Understand the basics of what is tissue engineering; including use of human cells, scaffolds and bioreactors

  • List clinical applications of tissue engineered products in regenerative medicine and understand where the field of research is today

  • Understand the role of scaffolds in tissue engineering included effect of surface, mechanical and biological properties

  • List different scaffold materials and understand the differences between them including for what application the materials should be used

  • Describe several different scaffold fabrication techniques such as electrospinning, solvent casting/particulate leaching etc and know at least three of them in detail

  • Understand the proliferation of cells and growth potential of primary cells

  • Describe how cells signal with each other and the ECM to determine the fate of a human cells

  • Understand the importance of apoptosis in multicellular development

  • Describe the early events that occur in tissue development from the first division in the egg to the migration of cells to form the different germlayers

  • Describe the formation of different organs due to gene regulations and cell signaling

  • Describe the different equipments used in cell culturing laboratory and list what are the crucial parameters controlling cell culture

  • Describe in what way and how the cells communicate with each other and with the surroundings

  • Describe the basic components of the ECM and its importance in tissue engineering

  • Understand why bioreactors are used in tissue engineering and give examples of in what applications they may be needed

  • In theory design a bioreactor with consideration to all the functions a bioreactor has to have to fulfill its role in tissue engineering


Practical objectives


Cell seeding laboratory



  • Perform the basics in cell culture; harvest, count cells and seed cells

  • Use the equipment present in a mammalian cell culture lab; pipettes, incubators, centrifuges, microscopes, flow hoods

  • Understand and be able to practice sterile techniques


Student project work



  • Plan and setup projects in tissue engineering including an understanding of the time aspects that are involved in tissue engineering projects

  • Perform scientific literature searches; perform article searches in databases and peer-reviewed journals

Innehåll

The following topics will be covered: Introduction Cell biology, the basis of growth and differentiation Growth factors In vitro control of tissue development Polymer Scaffolds Cell-polymer interactions Bioreactors

Organisation

The course will consist of 24 lectures and seminars and 40 hours of laboratory work. Laboratory work will be carried out in the project form where students will prepare the various scaffolds, characterize them, work with selected cells and study cell scaffold interactions in the bioreactor.
